Activation of Cilia-Independent Hedgehog/GLI1 Signaling as a Novel Concept for Neuroblastoma Therapy

    The study investigated a novel therapeutic approach for neuroblastoma (NB) by activating GLI1 signaling independently of primary cilia, which NB cells lack. Researchers identified an isoxazole (ISX) molecule capable of activating GLI1 without primary cilia, effectively blocking NB cell growth by removing the GLI3 repressor and inhibiting class I HDACs. This approach showed promise, particularly for high-risk NB patients with poor prognosis, as higher GLI1 expression correlated with better survival. The study suggested that small molecule-mediated GLI1 activation could be a promising strategy for NB therapy, offering a new avenue for treatment.
