The study developed a quantitative pipeline using fluorescently tagged histones and longitudinal imaging to observe chromatin compaction changes in live mouse epidermal stem cells. It revealed that chromatin compaction heterogeneity within the stem cell compartment was independent of the cell cycle and instead reflected differentiation status. Chromatin compaction transitioned gradually over days as cells exited the stem cell compartment. Additionally, dynamic transcriptional activity of Keratin-10, marking the onset of differentiation, preceded global chromatin compaction changes. This research provided insights into the relationship between chromatin architecture changes and transcription activation timing during stem cell differentiation.
