The study investigates the interactions between endothelial cells (ECs) and dermal papilla (DP) cells in hair regeneration and angiogenesis during aging. It reveals that these interactions decrease with age, leading to impaired hair regeneration and angiogenesis. Key signaling pathways, such as EDN1-EDNRA and CTF1-IL6ST, are crucial for these processes, with EDN1-EDNRA promoting taurine metabolism and hair regeneration, and CTF1-IL6ST enhancing alpha-linolenic acid metabolism and angiogenesis. The findings, supported by various experimental models including single-cell RNA sequencing and mouse organoid cultures, highlight the mutualistic relationship between hair follicles and blood vessels and suggest potential therapeutic targets for age-related hair loss.
