Efficacy of the Oral JAK1/JAK2 Inhibitor Deuruxolitinib in Adult Patients with Moderate to Severe Alopecia Areata: Pooled Results from the Multinational Double-Blind, Placebo-Controlled THRIVE-AA1 and THRIVE-AA2 Phase 3 Trials

    TLDR Deuruxolitinib significantly helps regrow scalp hair in adults with moderate to severe alopecia areata.
    The study combined results from two Phase 3 trials involving 1,209 adult patients with moderate to severe alopecia areata to assess the efficacy of the oral JAK1/JAK2 inhibitor deuruxolitinib. Participants received either a placebo, 8mg, or 12mg of deuruxolitinib twice daily for 24 weeks. The primary endpoint, a SALT score of ≤20 at Week 24, was achieved by 31.0% of patients in the 8mg group and 40.3% in the 12mg group, compared to 0.8% in the placebo group, showing significant efficacy (p<0.0001). Improvements were noted as early as 8 weeks, with 22.5% and 31.6% of the 8mg and 12mg groups, respectively, achieving a SALT score ≤10 at Week 24, versus 0% for placebo (p<0.0001). The study concluded that deuruxolitinib significantly promotes scalp hair regrowth in these patients.
