The study presents a novel treatment approach for alopecia areata (AA) using a ROS-responsive hydrogel to deliver miR-665, which targets STAT3 to reduce inflammation and promote hair follicle regeneration. The hydrogel, PVA-TSPBA, demonstrated excellent injectability and sustained release of miR-665, effectively counteracting IFN-γ-induced suppression in vitro and promoting hair regeneration in an AA mouse model. This biocompatible delivery system significantly restored follicular structure and reduced T-cell infiltration, offering a promising miRNA-biomaterial strategy for safe and durable AA treatment.
