Exogenous IL-27 Promotes Immunosuppressive T Cells and Prevents the Development of Alopecia Areata in C3H/HeJ Mice

    This study investigates the potential of IL-27 as a therapeutic option for alopecia areata (AA), an autoimmune disease causing hair loss. Using a skin-graft induction model in C3H/HeJ mice, researchers administered an adeno-associated virus expressing IL-27 (AAV-IL27). The treatment led to the generation of NKG2D-expressing CD8 T cells, typically associated with AA, but paradoxically protected the mice from developing the disease. The protection was linked to a 7-fold increase in IL-10 producing CD4 T cells, suggesting that IL-27 promotes immunosuppressive T cells, preventing AA. These findings highlight IL-27's potential as a therapeutic agent for AA, warranting further investigation into its mechanisms and therapeutic utility.
