Immune-Driven Activation of Hair Follicle Stem Cells as a Translational Model for Future Cell-Based Therapy Development

    This study explores the role of immune-driven activation of hair follicle stem cells (HFSCs) as a potential model for developing cell-based therapies for hair regeneration, particularly in conditions like alopecia areata. Using a murine model with controlled immune stimulation via diphenylcyclopropenone (DPCP), researchers identified that macrophage-mediated signaling is crucial for HFSC activation and hair regeneration. The study demonstrated that immune stimulation leads to synchronized anagen initiation, expansion of Lgr5+/Sox9+ HFSCs, and activation of the Wnt/β-catenin pathway. Macrophages were found to accumulate near HFSC niches, and their depletion significantly reduced HFSC proliferation, highlighting their essential role in immune–stem cell interactions. This model provides a valuable platform for testing new regenerative strategies and developing advanced cell-based therapies for alopecia and similar disorders.
