Conducted through a Phase II clinical trial with hundreds of participants, including an interim analysis of 10 participants, the research found that tofacitinib effectively reduced skin pathology in conditions like alopecia areata, psoriasis, and atopic dermatitis. It also decreased interferon and cytokine scores and levels of pathogenic autoantibodies without causing significant immune suppression. The findings suggest that JAK inhibition is a promising strategy for treating autoimmune conditions in DS, although further research is needed to explore its broader effects on DS's developmental and clinical features.
