The study revealed that Ku80 served as a novel receptor for Thymosin β4 (Tβ4) in endothelial cells, mediating its intracellular activity and influencing the expression of the PAI-1 gene, which is crucial for cell migration and wound healing. The interaction between Tβ4 and Ku80 was essential for this process, as mutations affecting their binding significantly altered PAI-1 expression. This indicated that Ku80 played a key role in the intracellular signaling pathway of Tβ4, independent of its known function of G-actin sequestering.
