The study investigated the role of laminin-511 in melanocyte migration and differentiation in mice. Mice with a keratinocyte-specific deletion of the laminin γ1 chain (Lamc1EKO) exhibited delayed coat pigmentation due to impaired melanocyte migration and differentiation. The absence of laminin-511, a pro-migratory substrate, led to reduced expression of differentiation-specific melanocyte enzymes and mislocalization of melanocytes. Laminin-511 was crucial for activating the chemokine receptor CXCR4, necessary for SDF-1-mediated migration of melanocytes into hair follicles. The findings highlighted the dual role of laminin-511 in regulating melanocyte migration and differentiation by providing a migration substrate and activating CXCR4 expression.
