Leucine-rich repeats containing G protein-coupled receptor 4 (LGR4) is crucial in cancer progression and cancer stem cell (CSC) properties by regulating signaling pathways like Wnt/β-catenin and MEK/ERK. It is overexpressed in various cancers, including skin, thyroid, and breast cancers, promoting cell proliferation and metastasis. LGR4 enhances CSC self-renewal and contributes to therapeutic resistance, particularly in acute myeloid leukemia and breast cancer. Targeting LGR4 with inhibitors could offer new cancer treatment strategies by reducing CSC characteristics and overcoming drug resistance. However, further research is needed to fully understand LGR4's functions and therapeutic potential.
