This study investigated the role of myosin regulatory light chain (RLC) and non-muscle myosin II (NM2) in the physiological function of inner ear hair cells. Using immunostaining and inhibitors in bullfrog saccules and mice, the researchers found that NM2 and MLCK inhibitors reduce hair bundle stiffness, inhibit spontaneous oscillation, and increase the resting open probability of transduction channels. Additionally, MLCK inhibition elevates hearing thresholds in mice. The study concludes that NM2 and RLC phosphorylation are crucial for modulating hair cell function and maintaining normal hair bundle operation.
