ROCK Inhibitor Y-27632 Maintains the Propagation and Characteristics of Hair Follicle Stem Cells

    The study showed that the ROCK inhibitor Y-27632 effectively preserved the propagation and characteristics of hair follicle stem cells (HFSCs), maintaining key markers and a stable ratio over multiple passages. In contrast, HFSCs cultured with epidermal growth factor lost these characteristics. Y-27632-derived HFSCs could form spherical clusters, differentiate into keratinocytes and adipose cells, and regenerate hair follicles in vivo when combined with fetal dermal cells in immune-deficient mice. These results indicated that Y-27632 maintained the self-renewal and differentiation potential of HFSCs, highlighting their potential for tissue engineering and clinical applications.
