Sacituzumab Tirumotecan, a TROP2-targeting antibody-drug conjugate, is showing promise in treating gynecologic malignancies such as cervical, ovarian, and endometrial cancers. Preliminary Phase III trial results indicate a manageable toxicity profile and promising antitumor activity, especially in cervical cancer, with a 57.9% objective response rate when combined with pembrolizumab. In a Phase I/II study of 40 patients with advanced ovarian cancer, the objective response rate was 40%, with a median progression-free survival of 6.0 months. Endometrial cancer trials showed objective response rates of 29.8% and 34.1% in different dosage groups. Common adverse events include hematologic toxicity, oral mucositis, and alopecia. The ongoing trials aim to further assess its efficacy and safety, potentially offering new therapeutic options for difficult-to-treat gynecologic cancers.
