TLDR A unified framework is needed to improve skin organoid construction for research and clinical use.
This review discusses the construction strategies of skin organoids, emphasizing the need for a unified framework that links construction methods with developmental outcomes and experimental value. It identifies key determinants such as cell source, assembly methods, extracellular matrix, signaling control, and quality assessment, which influence the developmental complexity, reproducibility, and scalability of organoids. The review highlights advances in engineering, like air-liquid interface culture and 3D bioprinting, for enhancing tissue structure and function. It provides stage-specific evaluation and optimization strategies to establish standardized protocols, aiming to develop physiologically relevant skin organoid systems for research and clinical applications.
