This study optimized the extraction of TSG-enriched Polygonum multiflorum extract (PME) and evaluated its effectiveness against androgenetic alopecia (AGA) through a dual mechanism: inhibiting 5α-reductase by 92.7% and activating the Wnt/β-catenin signaling pathway. PME's efficacy was demonstrated in a testosterone-induced AGA mouse model, where it accelerated hair regrowth and increased hair follicle size and number. The study suggests that PME could be a promising natural therapeutic for AGA, offering a balanced approach by preventing follicular degeneration and promoting regeneration, potentially surpassing the efficacy of finasteride and minoxidil.
