Wnt Signaling Influences the Development of Murine Epidermal Langerhans Cells

TLDR Wnt signaling affects the development and growth of Langerhans cells in mice.
The study examined the influence of Wnt signaling on the development of murine epidermal Langerhans cells (LCs). It was observed that Wnt3A increased the accumulation of EpCAM+ dendritic cells by 33% in vitro, while the Wnt antagonist Dkk1 reduced their number by 21%. In vivo, Dkk1 expression in transgenic mice led to a 40% reduction in LC proliferation and a 26% decrease in LC densities by postnatal day 14. These results indicated that Wnt signaling affected LC development, proliferation, and phenotype, although it was not essential for LC development after weaning. The study suggested that Wnt signaling might be more crucial during earlier developmental stages, and further research was recommended to investigate the regulation of LC precursors in fetal mouse epidermis.
