The CMECG system enhanced wound closure efficiency to 90% compared to 70% in the control group by day 14. The system facilitated fibroblast and mammary epithelial cell proliferation and migration, restored skin layer continuity, and promoted collagen synthesis and remodeling. The healing effects are linked to miRNAs within CiMECs-EVs, which regulate cell proliferation, migration, angiogenesis, and inflammation, involving pathways like MAPK, PI3K-Akt, and TGF-β. This novel EV-chitosan hydrogel system offers a promising therapeutic strategy for clinical skin wound repair.
