February 2025 in “Cell Cycle” The study investigates the use of hair-follicle-associated pluripotent (HAP) stem cell sheets to enhance wound healing and reduce scarring in a mouse model. HAP stem cells, derived from the bulge area of hair follicles, can differentiate into various cell types, including keratinocytes and endothelial cells. When implanted into skin ulcers in C57BL/6J mice, these stem cell sheets significantly accelerated wound closure and reduced scar formation compared to non-implanted controls. The study also observed decreased thickness of the epidermis and dermis, reduced infiltration of M2 macrophages and myofibroblasts, and lower expression levels of TGF-β1, COL1A2, and COL3A1 mRNA in treated mice. These findings suggest that HAP stem cell sheets could be a promising clinical strategy for improving wound healing and minimizing scarring.
