April 2025 in “Science Advances” This study explores a novel silicate-based therapy for inflammatory dilated cardiomyopathy (iDCM), a severe condition resulting from myocarditis. Researchers developed composite microneedles and ion solutions using calcium silicate bioceramics to deliver SiO3 2− ions directly to myocardial tissue or through systemic circulation. These ions modulate the cell microenvironment by regulating CD4 + T/T helper 17 (T H 17) cells and their interactions with neutrophils and fibroblasts via the FOXO signaling pathway. The therapy inhibits the hyperdifferentiation of CD4 + T cells to T H 17 cells and the transformation of neutrophils and fibroblasts, disrupting the cycle of inflammation and fibrotic lesions in iDCM. This biomaterial-based strategy shows promise for treating iDCM.
