The document discussed the shift in pharmaceutical strategy around 2000, when British pharmaceutical companies moved from traditional drug discovery methods to evidence-based drug development, marking the beginning of Structure Guided Drug Development (SGDD). Despite this shift, the success rate of drug development remained low, with high costs and many lead compounds discarded due to adverse effects. Structural analysis of target proteins and ligand complexes was highlighted as crucial for designing better drug compounds, leading to the concept of "Drug Rescuing." Efforts were made to develop basic technologies for this strategy. Additionally, software for computer-aided drug design, such as myPresto, was developed to aid in drug design processes, featuring tools for molecular dynamics simulation and virtual screening. Challenges in CADD tools, such as handling halogen atoms and metal ions, were also discussed.
