The study developed a robust and validated chromatographic method for the simultaneous quantification of Gemcitabine (GEM) and Olaparib (OLA) in pancreatic tissue, crucial for evaluating drug delivery systems targeting pancreatic cancer. The method demonstrated high specificity, selectivity, and sensitivity, with recovery rates between 95% and 105% and a linear range of 0.5–10 µg/mL. It maintained stability over 30 days and met international guidelines for precision, accuracy, and robustness. This method supports the development of advanced formulations for pancreatic cancer treatment by enabling accurate assessment of drug penetration, distribution, and retention in pancreatic tissue.
