Matrix-Free Cryo-Microneedles Array Patch for 3D Hair Follicle Organoids Delivery and Rapid Hair Regeneration

    This study introduces a cryo-microneedles array patch (cryo-MAP) technique for transplanting 3D hair follicle organoids, which are cultured without a matrix or serum and preserved with low-DMSO cryopreservation. The method avoids growth suppression from hydrogels, allowing hair follicles to germinate within 3 days. Proteomics analysis showed that cryopreservation maintained high cell viability and enhanced the organoids' differentiation capacity. The transplanted organoids successfully regenerated complex skin structures, achieving hair growth through the skin in about 15 days with a success rate of over 86%. This advancement presents new opportunities for organoid transplantation in tissue engineering and regenerative medicine.
