Despite mild phenotypes in loricrin knockout mice, backup mechanisms preserved quasi-homeostasis, highlighting the epidermal barrier's role in adaptive responses. The research emphasized sulfur metabolism's influence on immune responses, with the KEAP1/NRF2 pathway being vital for keratinization and stress responses. The absence of LOR led to structural fragility but not impaired permeability, suggesting compensatory mechanisms. The study also explored the role of vitamins A and D in immune regulation and keratinization, concluding that metabolic cues, such as retinoic acids, are significant for maintaining immune functions and tissue homeostasis. Further research was deemed necessary to fully understand these mechanisms.
