Single-Cell RNA Sequencing Highlights the Role of Proinflammatory Fibroblasts, Vascular Endothelial Cells, and Immune Cells in the Keloid Immune Microenvironment

    The study utilized single-cell RNA sequencing to investigate the immune microenvironment of keloids, focusing on proinflammatory fibroblasts (piF) and vascular endothelial cells (VEC). Analysis of three keloid lesions and two healthy skin samples revealed that piF and VEC are significantly elevated in keloid tissue. These cells are implicated in shaping the keloid microenvironment through increased expression of immune-associated genes and decreased expression of inflammatory chemokines. Additionally, VECs may act as a conditional immune subpopulation due to the upregulation of immune suppressive genes. The study also identified macrophages and Tregs as key players in the keloid immune landscape, suggesting potential therapeutic targets for keloid treatment.
