Joint Public Review: TLR2 Regulates Hair Follicle Cycle and Regeneration via BMP Signaling

The study reveals that TLR2 plays a crucial role in maintaining hair follicle homeostasis and regeneration by regulating hair follicle stem cells (HFSCs) through BMP signaling. TLR2 is expressed in a cycle-dependent manner in healthy hair follicles, and its activation is essential for countering inhibitory signals. In conditions like aging and obesity, both TLR2 and its ligand, carboxyethylpyrrole (CEP), decrease, leading to reduced hair growth. However, administering CEP can stimulate hair regeneration via a TLR2-dependent mechanism. This research highlights the importance of TLR2-mediated innate immunity in HFSC activation and suggests potential therapeutic strategies for preventing hair loss.
