Researchers conducted single-cell RNA sequencing on affected scalp biopsies from both human AA patients and C3H/HeJ mice, revealing increased expression of ITK and LAT in T cell clusters, suggesting a role for the TFK pathway in AA. In vivo experiments with C3H/HeJ mice showed that the dual JAK3/TEC inhibitor ritlecitinib effectively prevented and reversed AA, while the ITK inhibitor BMS-509744 had a partial effect. Other inhibitors tested did not show efficacy. These findings suggest that ITK inhibitors could be promising therapeutic targets for AA and potentially other autoimmune disorders.
