Dupilumab for Prurigo Nodularis: Real-World Outcomes Up to 104 Weeks From the Dupilumab Italian Prurigo Nodularis Study

The Dupilumab Italian Prurigo Nodularis (DUPItaPN) study assessed the long-term effectiveness and safety of dupilumab in 543 adults with prurigo nodularis over 104 weeks. The study demonstrated significant improvements in itch severity, skin lesions, sleep, pain, and quality of life, with a notable decrease in the Worst Itch Numeric Rating Scale score from 8.69 at baseline to 1.72 at week 104. A clinically meaningful itch reduction was maintained by 86.9% of patients, and 81.1% achieved an Investigator Global Assessment for PN-Stage score of 0 or 1. Dupilumab was well tolerated, with only 2.9% discontinuing due to adverse events, and no cancer progression or viral reactivation was observed. The findings support dupilumab as a long-term treatment option for moderate-to-severe PN, effective even in older adults and those with comorbidities.
