Fibronectin Meshwork Controls Epithelial Stem Cell Fate

    The study demonstrated that fibronectin played a crucial role in hair follicle stem cell (HFSC) fate and tissue regeneration. It was specifically enriched in HFSCs at the start of regeneration. When fibronectin was conditionally deleted in the HFSC compartment, it resulted in a blockade of hair regeneration and impaired stem cell location and fate. However, these issues were resolved with the dermal injection of exogenous fibronectin. The research further revealed that SLC3A2, the main integrin coreceptor, was essential for the integrin-dependent assembly of the fibronectin matrix, acting as a molecular relay for niche signals. This fibronectin-integrin-SLC3A2 cascade was found to finely tune HFSC fate and enhance tissue regenerative power.
