Nanoparticle-loaded dissolving microneedles (DMNs) offer significant potential for enhancing transdermal drug delivery by overcoming the stratum corneum barrier, combining the advantages of high payload and controlled release. Despite their promise in precision medicine, no such products have been approved due to a lack of understanding of their in vivo fate, which is a major obstacle to clinical translation. This review highlights the current research status and emphasizes the need to investigate the in vivo fate of these DMNs to advance their development and application.
