The document reviewed the advancements in engineered polypeptide-based drug delivery systems (EPP-DDS) for cancer therapy, highlighting their high biocompatibility, biodegradability, and ability to target specific sites, such as overcoming the blood-brain barrier. It summarized recent developments in manufacturing polypeptides and preparing EPP-DDS using green biochemical engineering for various cancer treatments, including chemotherapy, immunotherapy, photodynamic therapy, gene therapy, and combination therapy. The review also discussed inflammation-mediated cancer treatment strategies with EPP-DDS and emphasized the need for innovation in green biochemical engineering for the customized design and production of polypeptides, noting the potential of EPP-DDS to improve therapeutic efficacy and reduce side effects in clinical settings.
