Multiomic Characterization of Treatment Response to JAK Inhibition in Patients with APECED

The study investigates the effects of JAK1/2 inhibitor ruxolitinib on autoimmune responses in four APECED patients, revealing that the treatment reduces IFNγ-driven inflammation and normalizes tissue homeostasis, as evidenced by hair regrowth. The research highlights the multifaceted interactions between immune and stromal cells and the downregulation of pro-inflammatory responses. While the treatment shows promise, it also presents potential side effects such as anemia and weight gain. An expanded analysis of 89 additional patients identified a subgroup with severe symptoms and distinct inflammatory profiles, emphasizing the need for a comprehensive understanding of JAK inhibitors' systemic and tissue-specific effects for safe therapy.
