Ritlecitinib for the Management of Alopecia Areata: A Narrative Review of Real-World Evidence and Selected Post Hoc Analyses

Ritlecitinib, a JAK3 and TEC kinase inhibitor, shows promise as an effective treatment for alopecia areata (AA), demonstrating significant hair regrowth and reduced disease severity in both clinical trials and real-world studies. The ALLEGRO phase 2b/3 trial with 522 patients highlighted its efficacy, although prior treatments like corticosteroids influenced early responses. Real-world evidence from Italy and China supports its effectiveness, especially in JAK inhibitor-naïve patients, with mild adverse events reported. Studies in adolescents and children also showed significant improvements. However, the literature is limited and heterogeneous, necessitating larger, long-term studies to confirm its long-term safety and effectiveness, particularly in diverse and pediatric populations.
