    The study presented a method to measure the solubility of various solid chemicals in the stratum corneum (SC) under different relative humidity (RH) conditions, which is crucial for predicting dermal drug delivery and assessing skin exposure to chemicals. This method distinguishes between dissolved molecules within the SC and those present on the skin surface, and it allows for the study of hydrophobic chemicals without organic solvents. By comparing solubility in SC and delipidized SC, the method estimates chemical distribution between extracellular lipids and corneocytes at different hydration levels. The study found that solubility in porcine SC varies with hydration, affecting molecular uptake and transport across the skin. This methodology could be applied to other heterogeneous materials with complex structures.
