June 2022 in “Journal of Orthopaedic Surgery and Research” This study investigated the effects of Wedelolactone (WDL), a compound from the Wedelia chinensis plant, on particle-induced osteolysis using a mice model with 32 subjects. The study found that bone mineral density (BMD) significantly improved in the 8-week WDL group compared to the vehicle group, although there was no significant difference in bone volume/tissue volume (BV/TV) and trabecular bone thickness (Tb.Th) between the 4-week and 8-week treatments. Additionally, osteoclast numbers were lower in the 4-week WDL group compared to the vehicle group. The findings suggested that oral administration of WDL could reduce the severity of osteolysis without adverse effects, indicating its potential as a promising alternative to pharmaceuticals for reducing osteolysis with fewer side effects. However, limitations included the use of a flat bone model and the need for further research to explore inflammatory markers and refine the treatment approach.
