September 2000 in “Cancer” The study investigated the toxicity profile and pharmacokinetics of Doxil in 45 patients with metastatic breast carcinoma, previously treated with chemotherapy. Six different dose schedules were tested, and pharmacokinetics were examined in 24 patients. Results showed that stomatitis and skin toxicity (PPE) were dose and schedule dependent, with higher doses and shorter intervals increasing severity. Myelosuppression was mild, and hair loss was infrequent. Cardiac toxicity was rare, occurring in only 1 patient. Doxil demonstrated significant antitumor activity, with objective responses in 9 patients. Pharmacokinetics showed a long half-life and linear increase in Cmax and AUC with dose. Toxicity correlated with pharmacokinetic parameters, suggesting that pharmacokinetic guidance could optimize Doxil dosing.
