March 2024 in “Non-coding RNA Research” This study investigates the role of DNA methylation in chemotherapeutic alopecia using a mouse model induced by cyclophosphamide (CTX). It was found that DNA methylation of microRNA-365-1 (miR-365-1) plays a significant role in the apoptosis of hair follicle stem cells by targeting the DAP3 gene. The study demonstrated that hypermethylation of miR-365-1 leads to increased apoptosis in hair follicle stem cells, as evidenced by reduced expression of hair follicle stem cell markers and increased apoptotic markers. Overexpression of miR-365-1 reduced apoptosis, but this effect was partially reversed by overexpressing DAP3. These findings highlight the importance of miR-365-1 methylation in the regulation of hair follicle stem cell apoptosis during chemotherapeutic alopecia.
