It was found that the absence of podoplanin in epidermal keratinocytes did not affect the early stages of tumor development, such as tumor initiation, size, differentiation, or malignant transformation rates. However, the lack of podoplanin reduced tumor cell invasion, particularly single cell invasion, and decreased peritumoral lymphangiogenesis. These findings suggested that while keratinocyte-expressed podoplanin was not necessary for the initial stages of skin cancer development, it played a role in the progression of established tumors.
