August 2026 in “Scientific Reports” This study explores the effects of an oil-soluble extract from Lentinula edodes (Shiitake), standardized for ergosterol content, on skin health. The extract, SMosE, was shown to enhance vitamin D receptor-dependent transcription in human keratinocytes and dermal fibroblasts, promoting wound repair and improving skin barrier function. Ex vivo experiments confirmed increased levels of cytokeratin 10 and claudin-1, indicating enhanced epidermal maturation. A clinical trial with 39 adults demonstrated that SMosE improved skin hydration, reduced transepidermal water loss, and regulated sebum levels after 28 days of use. These findings suggest that SMosE could be a promising ingredient for dermatological and cosmetic applications.
