Researchers synthesized spermine-derived ionizable lipids with varying alkyl chains and formulated them into Sper-N/siRNA lipid nanoparticles. In vitro experiments showed that Sper-12-based nanoparticles effectively delivered siRNA for AR gene silencing. In vivo studies on AGA model mice demonstrated that intradermal administration of Sper-12/siAR nanoparticles suppressed aberrant AR protein, promoted hair follicle proliferation, and alleviated hair loss symptoms. This suggests that Sper-12/siRNA nanoparticles could be a promising platform for siRNA-based AGA treatments.
