The HPF demonstrated significant antioxidant activity and improved wound healing outcomes compared to other fractions and a non-treated group. It reduced pro-inflammatory cytokines IL-1β and TNF-α, increased TGF-β1 levels, and promoted collagen production and remodeling, which are crucial for preventing scar formation. Additionally, HPF enhanced the development of hair follicles and sebaceous glands. Molecular analyses showed that HPF upregulated TGF-β1 and downregulated Smad3, contributing to its wound healing efficacy.
Alopecia involves complex disruptions in immune, oxidative, apoptotic, and regenerative pathways within the hair follicle microenvironment. This review highlights the potential of a biomarker-guided therapeutic map to improve treatment precision by focusing on six key biomarkers: TNF-α, IL-6, MDA, caspase-3, VEGF, and β-catenin. Analysis of 15 animal studies from 2014 to 2024 revealed that TNF-α and IL-6 are linked to inflammation and immune damage, MDA to oxidative stress, and caspase-3 to apoptosis. Conversely, VEGF and β-catenin, which support angiogenesis and regeneration, are suppressed in alopecia but can be restored through effective treatments. Therapeutic strategies such as botanical extracts, bioactive compounds, and advanced delivery systems have shown promise in reducing harmful biomarkers and enhancing regenerative pathways. This biomarker-centered approach offers a promising framework for developing targeted, multitargeted alopecia treatments.
